Luminex
Presentation
The Luminex platform of the IMRB is located on the 1st floor of the Research building of the Henri Mondor hospital (Access map to the platform).
It is equipped with a Bio-Plex 200 device that, thanks to Luminex xMAP technology, can detect and quantify from 25 µl of sample to 100 different analytes. The platform is equipped with all the necessary equipment to perform complete assays according to the protocols of the different Luminex kit suppliers.
The equipment as well as the personnel of this platform are provided by the immunomonitoring platform of the Physiopathology and Immunotherapies in HIV Infection team (team Lelièvre) of the IMRB and the Vaccine Research Institute. Since the creation of the Luminex platform in 2011, more than 75 complete assays have been performed.

Materials available
- The Bio-Plex Washstation can be washed with Luminex kits using both polystyrene beads and magnetic beads.
- The Bio-Plex 200 is an open system allowing the use of several types of assay kits (Bio-Rad, Merck-Millipore, bio-techne, Affymetrix) and samples (plasma, serum, tissue lysate, culture supernatant, secretions) from several species (human, mouse, rat, monkey, dog, cat, pig).
The Bio-Plex 200 is an analyzer built around three technologies:
– The first is the family of fluorescent microspheres (or beads). These are labeled with different ratios of 2 fluorophores with different emission spectra in order to assign a unique spectral signature to each bead. Each set of beads is then conjugated to a capture molecule (enzyme/substrates, DNA, receptors, antigens, antibodies).
– The second is a flow cytometer equipped with 2 lasers and all the associated optics to measure the biochemical reactions that take place on the surface of the microspheres. In order to detect and quantify each analyte, a detection molecule specifically recognizing the analyte and a fluorescent reporter molecule are added. The red laser then excites the fluorescent markers of each bead, and the green laser excites the fluorescent reporter molecule associated with the bead, allowing the quantification of the captured analyte.
– The third is a high-speed digital signal processor for transforming fluorescent signals into data (Fluorescence intensities and concentrations).
The Bio-plex Washstation as well as the Bio-Plex 200 are under maintenance contract with the company Bio-Rad. They are checked and maintained every year.
The pipettes used by the platform personnel are also checked every year.
The platform staff has been trained to work with this Luminex technology and more specifically with the Bio-Plex 200 analyzer, and is competent and has expertise. The Bio-Plex 200 is under the responsibility of the technical manager, who alone is authorized to perform the manipulations.

Examples of services provided
- Conducting 29-plex and 27-plex Luminex assays in patients with macrophage myofasciitis (MMF) [Cadusseau et al, Curr Med Chem, 2014] and chronic obstructive pulmonary disease (COPD) [Boyer et al, PLoS One, 2015].
